Products from the photolysis of diazoacetyl chymotrypsin.

چکیده
1. A new and convenient synthesis of p-nitrophenyl diazoacetate has been devised from p-nitrophenyl chloroformate and diazomethane. 2. p-Nitrophenyl diazoacetate reacts stoichiometrically with chymotrypsin to yield diazoacetyl chymotrypsin. 3. Photolysis of the diazoacetyl enzyme with light of wave length greater than 320 rnp yields a mixture of (a) glycolic acid and regenerated chymotrypsin and (5) modified protein which retains the carbon atoms of the diazoacetyl group. 4. Acid hydrolysis of the protein obtained from photolysis yields 0-carboxymethylserine, identified by chromatographic and other comparisons with an authentic sample and with material formed on photolysis of azaserine. 5. In addition, small amounts of 1 -carboxymethylhistidine and of 0-carboxymethyltyrosine were obtained. The origin of the former is uncertain, but the latter may arise from photolysis of a dimer of the diazoacetylated enzyme. 6. Most of the products presumably arise from a carbene formed as a primary product of the photolysis of the diazoester.

Xarboxymethylcysteine from the Photolysis
Photolysis of ethyl diazomalonyl trypsin and of ethyl diazomalonyl chymotrypsin, followed by hydrolysis, yields several products. One of the major products has now been identified as S-carboxymethylcysteine. Since neither trypsin nor chymotrypsin contains an -SH bond, the reaction must involve cleavage of a disulfide bond by the carbene generated on photolysis. The products so far identzed from...
متن کاملChymotrypsin activity extracted from beluga (Huso huso) intestine under some physical and chemical factors
In the present study, the physical and chemical factors including optimum temperature and pH, the effect of inhibitors, metal ions, oxidizing agents, and surfactants were assayed on the chymotrypsin activity from beluga (Huso huso) intestine. According to the obtained results, the optimum temperature and pH of the chymotrypsin were recorded at 50°C and 8, respectively, and its activity was extr...
متن کاملComparing the Efficiency of UV/TiO2 and UV/O3 Processes in Degradation of Sulfonamide Antibiotics in Aquatic Solution
Background and purpose: Emerging pollutants such as antibiotics are resistant to biodegradation. The aim of this study was to compare the effect of photocatalytic and Ozonation photolysis on decomposition of Sulfonamide antibiotics (Sulfacetamide, Sulfathiazole, Sulfamethoxazole, and Sulfadiazine) in aquatic environments. Materials and methods: In this study, experiments were conducted discont...
متن کاملPhotodegradation of Methylene Blue Solution via Au Doped TiO2 Nanocomposite Catalysts Prepared Using Novel Photolysis Method
Gold doped TiO2 has been successfully synthesized via the photolysis method and is characterized by different techniques. NPs of gold doped TiO2 were utilized for the degradation of methylene blue as a material pigmentation pollutant. The substitution of Au on TiO2 surface was established via XRD, EDX, TEM, and FT-IR techniques. The TEM and SEM results appea...
متن کاملDirect detection and reactivity of the short-lived phenyloxenium ion.
Photolysis of protonated phenylhydroxylamine was studied using product analysis, trapping experiments, and laser flash photolysis experiments (UV-vis and TR(3) detection) ranging from the femtosecond to the microsecond time scale. We find that the excited state of the photoprecursor is followed by two species: a longer-lived transient (150 ns) that we assign to the phenoxy radical and a shorter...
